Gene Delivery by Electroporation
Motivation: Genes could be used to correct skin-specific disorders as well as for the
production of proteins secreted into the skin and circulatory system. For example,
fibroblasts in the skin may secrete protein factors to treat systemic conditions such as
Demonstration: lacZ gene was electroporated into mouse skin. The lacZ gene
expresses b-galactosidase which hydrolyzes 5-Br-4-Cl-3-indolyl-b-D-galactoside
to indigo (blue dye). Observation of the blue stain thus indicates gene expression.
The effects of applied pressure and voltage pulses on depth of expression and
number of transfected cells were studied.
