Fourier-transform infrared (FTIR) spectroscopy
has been proposed for nearly 25 years and yet
has not gained acceptance among the biological
community. Most secondary structure determination
is performed by ultraviolet circular dichroism (UVCD)
spectroscopy. The sample preparation and relatively
large amount of sample required are cited as
disadvantages. We have explored the use of
single-pass ATR-FTIR as an alternative method.
