The analysis of protein spectra obtained by single-pass
ATR-FTIR is based on the acquisition of a spectral library.
We have studied 25 proteins and use principal component
regression (PCR) methods. The prediction of secondary
structure is quite accurate using this method. The amount
of sample required is small and the measurement time is
several minutes. The disadvantage at present is the lack of
control over the hydration state of the sample. This is
being remedied using a humidity chamber. Control
experiments show that the proteins are not being denatured
under the conditions of the experiment. However, as
discussed below, further experiments are underway to
create biocompatible tips and to bind proteins through
particular interactions such polyhistidine tags.
